    Effects of Chitosan on Seed Germination of Hyacinth Bean( Lablab purpureus(Linn.)Sweet)Under Salt Stress

    Hyacinth bean cultivar "No.1Chunbian" was used as the test material. Its seeds were soaked with different concentration of chitosan solution(50,100,125,150,175,200,250 mg/L),and then treated by 6 g/L NaCl solution during seed germination. The effects of different concentrations of chitosan on hyacinth bean seed germination and seedling physiological indices under salt stress were studied. The results showed that under the salt stress,the seed germination potential,seed germination rate,seed germination index and the root length,shoot length,activity of SOD in the seedling had the trend of increase and then decrease with the increase of the concentration of chitosan,and the tested indices reached the maximum value at 150 mg/L chitosan,but MDA content reached the minimum at 150 mg/L chitosan. Therefore,appropriate concentration of chitosan can alleviate salt stress to the lentils and the optimum treatment concentration of chitosan is 150 mg/L.

    In this paper,the author identified one particular small heat shock protein(sHSP)( shsp27 ,GenBank number FK669241.1) which contained a typical α- crystalline central domain from Callosobruchus maculatus with bioinformatics method. Then,the author used real- time quantitative PCR(RT-qPCR)to detect the mRNA relative expression levels of shsp27 in 4th larva of C. maculatus ,after being exposured to different temperature stress(4,15,30,37 ℃,and 4 ℃ 24 h then back to 30 ℃ treatments(Rec)). The results showed that the low temperature(4,15 ℃)and high temperature(37 ℃)heat stress up-regulated the relative expression of shsp27 ,and the high temperature stress upregulated it stronger,that of Rec was slightly higher than that of CK,but not significantly. It is considered that there is no correlationship between shsp27 expression and high/low temperature adaptability.

    In this paper,the processes of extraction,separation and purification of Phaseolus vulgaris agglutinin(PHA)from the common bean "Bean Saint esprit a oeil rouge" fresh pods were optimized. The effects of extracting agent,material-liquid ratio,pH and extracting time on PHA were studied,and the combined method of ammonium sulfate precipitation, DEAE- 52 anion exchange chromatography and Superdex-200 gel filtration chromatography for extracting,isolating and purifing the PHA from fresh pods of common bean has been established. The main results were as follow:1)The optimized technical parameters for common bean extraction process were material-liquid ratio 1∶25,pH 6-8,extracting time 8 h,PBS as extracting agent;2)The high purity PHA was obtained with ammonium sulfate precipitation,dialysis,DEAE-52 anion exchange chromatography,concentrated by ultrafiltration and Superdex-200 gel filtration chromatography. The obtained PHA shows one band with electrophoresis test and has blood clotting activity.

    Fresh pods of common bean contain a variety of anti-nutritional factors,lectin is one of the main endogenous toxins. It can cause poisoning phenomena while improper eating. In this study,the contents of lectin in fresh pods were detected in 121 selected common bean cultivars,the genetic variations were analysed,the screening range was enlarged with extreme low and extreme high lectin content. The results revealed significant difference of lectin in the tested cultivar population(P < 0. 01),its coefficients of variation( CV )was 98. 34%. Meanwhile,contents of lectin in all cultivars ranged from 0. 005 mg/g to 1. 330 mg/g,and the mean value was 0. 412 mg/g. The cultivar frequency distribution curve showed a main peak and nearly normal distribution,but slightly long on dual-tails,which demonstrated the distribution of cultivars with extreme low or high level on lectin. Therefore,three cultivars containing less lectin were identified. It will provide experimental basis and data support for genetic improvement and rational utilization of non endogenous toxicity germplasm.

    Cowpea rust,a fungal disease caused by Uromyces vignae Barcl,is one of the major cowpea diseases worldwide. The main targets of the pathogen are leaves,the petioles and pods may be damaged in severe cases. The disease can dramatically influence the cowpea production by reducing the yields and the quality. In this paper, researching progresses on cowpea rust are reviewed, including pathogenic characteristics,disease epidemics,resistant germplasm resource evaluation and breeding,physiological and biochemical mechanism of plant resistance, and molecular genetics of rust resistance,also the author discusses the further direction in resolving this disease.

    With different digestion method and measuring method,the cadmium content of harmful inorganic elements in foods was determined and compared,meanwhile,the statistical methods for data processing were used, and then the data were analyzed, thus the more suitable digestion and determination methods were found. The results showed that wet digestion with graphite furnace atomic absorption spectrometry and the wet digestion with flame atomic absorption spectrometry were used to measure the recovery,the differences of recovery rates were small,the standard deviation of wet digestion with flame atomic absorption spectrometry was the smallest and the precision was the highest.Comprehensive comparison showed the determination of cadmium in foods with the wet digestion and flame atomic absorption spectrometry was most reasonable.

    In this paper,volatile flavor compounds of Artemisia selengensis leaves and Artemisia selengensis tea were extracted with ethanol as the polar solvent,the compounds were comparatively analyzed with GC-MS technology,and the relative content of each component was calculated with area normalization. The results suggested that there were 41 kinds of volatile constituents in Artemisia selengensis leaves,mainly including the alkane(9 species)and the olefin(19 species),the contents were 47. 22% and 35. 52% respectively. There were 28 kinds of volatile constituents in Artemisia selengensis tea, mainly including the alkane(13 species)and the ester compounds(4 species),and the contents were 60. 70% and 25. 32% respectively.

    Lysine acetylation of histone plays an important role in regulation of gene expression in plant cells,meanwhile it affects the enzyme activity of glycolysis,tricarboxylic acid cycle and photosynthetic pathway. Therefore,it is of great significance to identify histone acetyltransferase(HAT)and histone deacetylase(HDAC)of metabolism regulating enzymes. On the contrary,metabolites such as acetyl-CoA and NAD+ are involved in processes of protein acetylation and deacetylation,their intracellular levels may influence on activity of HAT and HDAC. This paper illustrates the role of HAT,HDAC and their research progress from protein lysine acetylation process,classification and cell distribution,and metabolic enzyme activity.

    Towards the demand of intelligent iterative development of the smart home system at present,this paper analyzes the current researches on technical realization of smart home system control,proposes the method of design and implementation of smart home system control based on Python Django. With Python Django,the designs of system structure,communication protocols,gateway system are realized. Combined with the CAN network,this paper realizes complete application of smart home system. Through the scalability analysis of security module of the smart home system,it is illustrated that Python Django is more suitable for rapid iteration development of smart home system.

    A new idea with GM(1,1)model to predict the change of target feature is proposed in this paper. The method divides the target area into blocks,and calculates the sum of the pixel in the block area. At the same time,the short time series are used to cumulativly process the pixels sum sequence to generate a new sequence. The target prediction model is obtained with GM(1,1)model. GM(1,1) pixel prediction model has a strong predictive ability for the target,and has a better predictive ability for the short-term characteristics change. In this paper,a new effective tracking algorithm is proposed,which combines the feature change with the prediction. After testing the proposed method,it is found that the tracking performance is improved significantly compared with the traditional tracking algorithm.

    The dynamic response of a temporary support structure of a highway tunnel was analyzed in this article. The dynamic response at different conditions of the temporary support structure,composed of a skeleton and concrete lining structure ,was analyzed by ANSYS. With simplifying it as single concrete material shell supporting structure model,it completed the preparatory work of the dynamic analysis of the whole tunnel model under blasting load. It was found that the natural frequency of eachorder modal was within the main band of blasting vibration wave when doing the modal analysis of finite element model,which provided the reference for the safety of the blasting construction of the tunnel.

Taking some roads of Xiamen University of Technology as the object,based on FARO Focus3D three dimensional laser scanner,the author carried out point cloud data collecting,projecting picture extracting,fast drawing of digital line graphic(DLG),to realize 2D information extraction and 3D model construction,meanwhile,the author compared the data with traditional total station data,and obtained the integrity and effectiveness of information of laser cloud data.

    Through a total of 1 680 students from 9 universities in Anhui province,the author used three scale of leisure participation, leisure satisfaction and subjective well- being to carry out the questionnaire survey,and combined with the relevant literature,this study found:there were canonical correlations in leisure participation,leisure satisfaction and subjective well-being. When the leisure participation frequency was high,or with a higher degree of leisure satisfaction,there was a higher degree of satisfaction to the overall quality of life,and subjective well- being is the main index to predict the quality of life. According to the results of the study,the author gave out suggestions for college students′ leisure education and future research.
