Abstract: In this paper，we discuss the effects on microscopic observation for cytoskeleton from the following five aspects：different experimental materials，type of extractive agent，extraction time，concentration of extractive agent Triton X-100 and fixation time of glutaraldehyde. The results show that：green Chinese onion，shallot，leek can substitute onion as cytoskeleton materials. When leek is used as cytoskeleton materials，the effect of 1%Triton X- 100 extraction for hybridprotein with the extraction time within 25-30 min is better；the concentration of extractive agent Triton X-100 within 1%-3% is better；the fixation time of glutaraldehyde within 15 - 25 min is better.
