Callus Induction Effect and Lectin Level of Common Bean

Abstract: The ′Bean Saint esprit a oeil rouge′ ′Bean White rice′ ′ 97-5 racks′ of common bean cultivars were chosen as raw materials. After seed germination,the cotyledons,stems and leaves of the beans were chosen as explants,respectively,and to culture them in MS medium with different concentrations of hormone(2,4-D,6-BA,KT)to observe the induction of callus and determine lectin level in callus. The results showed that the suitable medium of cotyledon,stem and leaf callus in the three common bean cultivars were MS + 3 mg/L 2,4-D + 0. 5 mg/L 6-BA + 0. 5 mg/L KT. The agglutination activity of plant lectin in the cotyledons and leaves callus of ′Bean Saint esprit a oeil rouge′ was the highest,it was 0. 27 HU. In conclusion,the cotyledons or leaves of ′Bean Saint esprit a oeil rouge′ were selected in medium of MS + 3 mg/L 2,4-D + 0. 5 mg/L 6-BA + 0. 5 mg/L KT which could produce the largest amount of lectin,it will provide the experimental basis and data support for further research of artificial production of bean lectin.
