Optimization of Extraction Process of Proanthocyanidins from ′Cabernet Sauvignon′ Residue of Ningxia by Response Surface Methodology

Abstract: Ethanol was used to extract the proanthocyanidins from Ningxia ' Cabernet Sauvignon' grape skin residue by adding cellulase and pectinase. Based on the single-factor experiment, the response surface methodology was used to investigate the effects of material-liqiud ratio, the amount of complex enzyme, extraction temperature, and extraction time on the yield of proanthocyanidins. Then,the quadratic polynomial regression model was established to analyze and predict the yield of proanthocyanidins. The best process conditions for proanthocyanidins are ethanol concentration 60%,compound enzyme addition 0. 4%,material-liqiud ratio 1∶20,pH value 4. 5,extraction temperature 60 ℃ ,extraction time 80 min. Results show that the yield of proanthocyanidins from Ningxia ' Cabernet Sauvignon' grape skin residue is 22. 914 mg/g.
