Identification and Value Analysis of lncRNA m6A Modification in Prognosis and Immune Microenvironment of Lung Cancer Patients

Abstract: Objective To determine the correlation and prognostic value of m6A-related lncRNA and immune infiltration in lung cancer. Methods The clinical-related information and RNA-Seq transcriptome data of lung cancer patients were downloaded from the TCGA database. Univariate Cox regression and Pearson analysis were used to screen out m6Arelated lncRNA. Consensus cluster analysis was used to divide the sample into two clusters, and LASSO analysis and minimum absolute shrinkage were used to construct a risk-scoring model. Results A total of 5 lncRNA expression profiles were screened,and lung cancer patients were divided into different subtypes. Cluster 2 had a better prognosis,and this cluster was related to immune cells. Five lncRNAs were selected to construct a risk model, and the prognosis of lung cancer patients in the high-risk group was poor. The expression of PD-L1 in tumors was significantly higher than in normal tissues. Univariate and multivariate Cox regression analysis showed that the overall survival rate was significantly related to stage and risk score,which could be used as an independent prognostic factor. The relationship between risk score and immune cell infiltration showed that immune cells were correlated with risk scores. Conclusion The results of this study indicate that m6A-related lncRNA may play an important role in the prognosis of lung cancer patients and the tumor immune microenvironment,which provides certain help to the treatment of lung cancer patients.
