Analysis of Dosing Pattern of Chinese Medicine Fumigation for Vertebral Artery Type Cervical Spondylosis Based on Data Mining Technology

Abstract: Objective To analyze the mechanism and efficacy of traditional Chinese medicine(TCM)compound preparation in treating obesity based on bioinformatics and efficacy study in vitro and provide the basis for the innovative application of commonly used TCM. Methods The active components and targets of TCM compound preparations were obtained by TCMSP. We screened the differential gene chip data by the GEO database to map the volcano. Cytoscape3.8.0 was used to construct the differential gene target map of TCM active ingredients and obesity,create a protein interaction network,and conduct an enrichment analysis of the GO and KEGG pathways. Finally ,MTT and Annexin V-FTIC/PI flow cytometry were used to verify cell survival and apoptotic effect. Results A total of 204 active ingredients were obtained in TCM compound preparation for treating obesity,and the main components were quercetin,kaempferol,luteolin,baicalein,etc. There were 28 obesity-related differential genes,and the PPI topological network of the differential genes was constructed. The GO and KEGG pathway was involved in the pathways related to insulin,hormone,lipid,and glucose metabolism. MTT confirmed that the drug had a significant concentration-time dependence on the preadipocytes of 3T3-L1 mice,and the IC50 value at 72 h was 2. 395 mg/mL. Moreover,it can induce the early apoptosis of preadipocytes. Conclusion This study provides important molecular targets,pathways,and mechanisms of action of TCM compound preparations in treating obesity through pharmacodynamics study in vitro,providing a basis for the clinical application of TCM.
