Effects of Exogenous Hormones on Proliferation and Rooting of a New Stevia Cultiver ′huinong No. 5′ in vitro

Abstract: In this experiment,MS was used as the basic medium and the stem segments of ′huinong No. 5′ stevia test-tube seedlings were used as experimental materials. The effects of the treatment combination of 6-BA,KT,Ad three exogenous hormone solutions at 6 concentrations and 0. 1 mg/L NAA on the proliferation and rooting of test-tube seedlings were studied. The results showed that the effects of adding an appropriate concentration of exogenous hormone were obvious. The medium suitable for the secondary culture of stevia test-tube seedlings was MS + 6-BA 1. 0 mg/L + NAA 0. 1 mg/L + sugar 30 g/L + AGAR powder 5. 5 g/L. The medium suitable for the proliferation of stevia test-tube seedlings was MS + 6-BA 2. 0 mg/L + NAA 0. 1 mg/L + sugar 30 g/L + AGAR powder 5. 5 g/L,the proliferation coefficient can reach 3. 50. The optimal rooting medium for test-tube seedlings was 1/2 MS + NAA 0. 1 mg/L + sugar 20 g/L + AGAR powder 5. 5 g/L. Different kinds of exogenous hormones,different concentrations and different combinations of treatments have different effects on the proliferation and rooting of stevia test-tube seedlings.
