Identification of Genetic Relationship Among Several Chinese Strains of Ganoderma with RAPD and rDNA Molecular Markers

Abstract: Uses several common Chinese strains of ganoderma as researching material,adopts RAPD,18SrDNA and ITS molecular markers to analyse the genetic relationship among each strain of ganoderma,and compares the analysing results of three methods,to explore the suitable technical method on analysing ganoderma. RAPD analysis show that six strains are clustered into 3 groups the similarity coefficient of 0. 584 Ganodermaluidun,Ganodermaluidum HG,Ganodermasinenses and Ganodermaatrum as a category;Xuezhi and Coriolusversicolor are each clustered into a single category. ITS analysis show that ganoderma has lower similarity(43. 8% ~ 88. 0%),similarity of Ganodermasinenses and Xuezhi is 43. 8%. In the lower similarity level,Ganodermaluidun and Ganodermasinenses are classified as a category,Coriolusversicolor and Xuezhi are each classified as a single category,which is consistent with the RAPD analysis and also consistent with the traditional classification. The results of 18SrDNA molecular markers are very different with the first two results,indicating that 18SrDNA molecular markers are not suitable for the classification research of ganoderma.
