菜豆子叶节诱导成苗技术初步研究

Preliminary Research of Common Bean Seedlings Induced Technology by Cotyledonary Node

摘要: 以“Bean white rice”菜豆为研究材料,研究了在MS 培养基上添加不同生长素IBA 和NAA 以及细胞分裂素6-BA 和KT 对菜豆子叶节诱导不定芽和根的影响,并对菜豆组培再生苗进行炼苗和移植。研究结果表明:最佳芽诱导培养基为MS + 0. 5 mg/L 6-BA + 0. 1 mg/L KT + 1. 0 mg/L IBA ,不发芽分化系数平均达到2. 89;最佳生根培养基为MS + 0. 1 mg/L IBA,生根率为100%;菜豆组培再生苗移栽后可以正常存活。

Abstract: The effects of different concentrations of auxins IBA(indole butyric acid), NAA(naphthalene acetic acid)and cytokines 6-BA(6-benzyl adenine),KT(kinetin)on the induction of adventitious buds and roots were studied from the cotyledonary node of "Bean white rice" common bean,and the common bean′s tissue culture seedlings were cultivated and transplanted. The results showed that the optimum medium for buds induction was MS + 1. 0 mg/L IBA + 0. 5 mg/L 6-BA + 0. 1 mg/L KT,the cofficient of the bud differentiation was 2. 89. The optimum rooting medium was MS + 0. 1 mg/L IBA,the rooting rate was 100%. The common bean′s tissue culture seedlings can survive after transplanting.
