戒毒一号的高效液相色谱指纹图谱研究

摘要: 目 的 建立中药复方戒毒一号的高效液相指纹图谱,为戒毒一号的质量控制提供依据。方 法 采用高效液相色谱(HPLC)法,Thermo 公司Ultimate TM XB-C18 色谱柱,延胡索甲素、延胡索乙素、小檗碱、甘草酸以乙腈-水-磷酸缓冲液为流动相,体积流量为1. 0 mL/min,柱温30℃,检测波长为280 nm;戒毒一号中的人参皂苷Re、人参皂苷Rb1以乙腈-纯水为流动相,二元梯度洗脱,体积流量为1. 0 mL/min,柱温30℃,检测波长203 nm。结 果 10批戒毒一号水煎剂中共有峰的相对保留时间RSD 小于1%。在选定的色谱条件下确定了19个峰为戒毒一号的特征指纹峰,10批样品平均相似度为0. 988。结 论 该法重现性好,可为戒毒一号的质量控制提供依据。

Abstract: Objective The aim of this study is to develop the fingerprints of the Jieduyihao(JDYH) with high performance liquid chromatographic (HPLC) analysis and to provide the basis for the quality control of JDYH. Methods The HPLC fingerprints were conducted with the Ultimate TM XBC18(250 mm × 4. 6 mm)column in gradient elution. The analyses of corydaline,tetrahydropalmatine,berberine and glycyrrhizic acid were conducted with the mobile phase consisted of acetonitrile-waterphosphoric acid,at column temperature 30 ℃,flow rate of 1. 0 mL/min and detective wavelengths of 280 nm. The analyses of ginsenoside Re and ginsenoside Rb1 were conducted with the mobile phase consisted of acetonitrile-water in gradient elution,at column temperature 30 ℃,flow rate of 1. 0 mL/min and detective wavelengths of 203 nm. Results The fingerprints of ten batches of JDYH decoction were established,which contained 19 common peaks,the similarities among different batches were 0. 998, with RSD < 1% . Conclusion The analyses of HPLC fingerprints have high sensitivity and good reproducibility,which can provide a basis for scientific evaluation and quality control of JDYH.
