腹壁和肠系膜移植法制作大鼠子宫内膜异位症模型效果比较

Endometrium Transplantation at Abdominal Wall and Intestinal Loop on Making Endometriosis Rat Model

摘要: 目 的 比较不同移植部位建立大鼠子宫内膜异位症模型效果的差别,以期建立重复性高的子宫内膜异位症造模方法。方 法 SPF 级成年雌性SD大鼠11只,10%水合氯醛麻醉后,手术取出带有内膜、肌层、浆膜子宫组织,用可吸收线分别将其缝于左右腹壁各1块,左右肠系膜靠近肠袢处各1块。术后常规饲养4周后检测移植内膜存活情况及生长情况。结 果 开腹后见移植内膜体积明显增大,呈暗红色或透亮的小囊泡视为造模成功。术后大鼠死亡1只,存活大鼠腹壁移植造模成功率为65%,肠系膜移植造模成功率为95%,χ2检验P < 0. 05,两者差异有统计学意义。移植至腹壁的子宫内膜形成的囊肿体积(35. 53 ± 22. 58)mm3较肠系膜处(90. 29 ± 74. 93)mm3小,但两者比较差异无统计学意义。结 论 内膜移植至肠系膜更利于存活,且易形成较稳定的生长病灶,更利于子宫内膜异位症动物模型相关研究。

Abstract: Objective To compare the differences of endometrium transplantation at abdominal wall and intestinal loop on making endometriosis rat model,aimed to find a reliable method for endometriosisbased study. Methods 11 adult female SD rats were anesthetized by 10% chloral hydrate. Endometrium tissues were stitched to abdominal wall and mesentery with absorbable suture silk. Survival and growth of transplanted endometrium were checked four weeks later. Results The volume of grafts increased significantly,dark red or translucent vesicles indicated that the model was made successfully. 1 rat died within 1 week after surgery. 65% of grafts survived on the abdominal wall,which was significantly lower than transplanted on the mesentery(95%), P < 0. 05. There is no difference between the volume of endometrium on abdominal wall (35. 53 ± 22. 58)mm3 and mesentery (90. 29 ± 74. 93)mm3. Conclusion Endometrium transplanted to mesentery is beneficial for endometrium survival and growth,which can be used for endometriosis-based animal study.
