几种中国栽培灵芝品种的RAPD及rDNA分子标记鉴定

摘要: 以常见几种中国栽培灵芝菌株为研究材料,运用RAPD、18SrDNA和ITS 3种分子标记来分析各灵芝菌株之间的亲缘关系,同时比较3种分析结果,探索适于灵芝属研究的技术方法。RAPD分析结果表明,6种菌株在相似性系数为 0. 584的水平上聚为 3类:Ganodermaluidun、GanodermaluidumHG、Ganodermasinenses和 Ganodermaatrum为一类;Xuezhi和 Coriolusversicolor各单归为一类。ITS分析结果显示,灵芝菌株间相似性较低(43. 8 %~88. 0 %),其中Ganodermasinenses和Xuezhi的相似性为43. 8 %,在较低相似性水平上,Ganodermaluidun和Ganodermasinenses归为一类,Coriolusversicolor和Xuezhi各单归为一类,这与RAPD分析结果一致,也与传统分类结果基本一致。18SrDNA分子标记显示的结果与前两种分析结果有很大差异,说明18SrDNA分子标记并不适合灵芝属的分类研究。

Abstract: Uses several common Chinese strains of ganoderma as researching material,adopts RAPD,18SrDNA and ITS molecular markers to analyse the genetic relationship among each strain of ganoderma,and compares the analysing results of three methods,to explore the suitable technical method on analysing ganoderma. RAPD analysis show that six strains are clustered into 3 groups the similarity coefficient of 0. 584 Ganodermaluidun,Ganodermaluidum HG,Ganodermasinenses and Ganodermaatrum as a category;Xuezhi and Coriolusversicolor are each clustered into a single category. ITS analysis show that ganoderma has lower similarity(43. 8% ~ 88. 0%),similarity of Ganodermasinenses and Xuezhi is 43. 8%. In the lower similarity level,Ganodermaluidun and Ganodermasinenses are classified as a category,Coriolusversicolor and Xuezhi are each classified as a single category,which is consistent with the RAPD analysis and also consistent with the traditional classification. The results of 18SrDNA molecular markers are very different with the first two results,indicating that 18SrDNA molecular markers are not suitable for the classification research of ganoderma.
