葱白提取物对心肌梗死大鼠促血管新生作用及机制研究

Study on Angiogenesis Effect and Mechanism of Fistular Onion Bulb on Myocardial Infarction in Rats

摘要: 目 的探讨葱白提取物(fistular onion bulb,FOB)对促血管新生(angiogenesis)的作用 及其机制。方 法将 SD 大鼠,随机分成葱白提取物组、麝香保心丸组、模型组、假手术组,每组 10 只。在适应性喂养 1 周后,行左冠脉前降支结扎造模。CD34 免疫组化染色观察心肌梗死边缘 区微血管数量(microvessel quantity,MVQ)及微血管密度(microvessel density,MVD);ELISA 法 检测血清转化生长因子-β(transforming growth factor-β,TGF-β)含量;Western Blot 法检测大 鼠左室心肌组织丝裂原激活蛋白激酶(mitogen activated protein kinase,MEK)、细胞外信号调节 激酶(extracellular singnal-regulated kinase,ERK)蛋白含量;RT-PCR 法检测心肌梗死边缘组 织 MEK、ERK mRNA 表达量。结 果 与模型组相比,葱白提取物组、麝香保心丸组在 CD34 染 色后心肌梗死边缘区微血管计数大幅升高(P < 0. 01);葱白提取物组微血管计数低于麝香保心 丸 组(P < 0. 01)。 与 模 型 组 相 比 ,葱 白 提 取 物 组 、麝 香 保 心 丸 组 血 清 TGF- β 均 增 加(P < 0. 01,P < 0. 05)。 葱 白 提 取 物 组 及 麝 香 保 心 丸 组 MEK、ERK 蛋 白 表 达 量 均 增 加 ,MEK、ERK mRNA 扩增倍数显著升高(P < 0. 01);与麝香保心丸组相比,葱白提取物组 ERK 蛋白表达量、 ERK mRNA 扩增倍数均较低(P < 0. 01,P < 0. 05),MEK 蛋白表达量无明显差异(P > 0. 05)。 结 论葱白提取物具有促进梗死心肌血管新生作用,增加心肌梗死边缘区新生血管数目,其机 制可能与上调血清 TGF-β 水平及 MEK/ERK 信号通路表达有关。

Abstract: Objective To probe the function and mechanism of the fistular onion bulb(FOB)in promoting angiogenesis on myocardial infarction in rats. MethodsRandomly dividing the rats into 4 groups(10 in each group):the FOB group,the Shexiang Baoxin pill group,the model group,and the sham operation group. After one week of adaptive feeding,the model of left anterior descending coronary artery ligation was performed. The quantity and density of microvascular in the margin of myocardial infarction were observed by CD34 immunohistochemical staining. Serum TGF-β content was detected by ELISA. The protein levels of MEK and ERK in the left ventricular myocardium of rats were detected by Western Blot. The mRNA expression levels of MEK and ERK in the edge of myocardial infarction were detected by RT-PCR. ResultsCompared with the model group,The MVD value of CD34 staining in The FOB group and the Shexiang Baoxin pill group was significantly increased(P < 0. 01). The MVD value in The FOB group was lower than that in the Shexiang Baoxin pill group(P < 0. 01). Compared with the model group,the level of serum TGF-β in the FOB group was significantly increased(P < 0. 01),and the level of TGF-β in the Shexiang Baoxin pill group was also increased(P < 0. 05). The protein expression levels of MEK and ERK were increased,and the amplification factor of MEK and ERK mRNA was significantly increased (P < 0. 01). Compared with the Shexiang Baoxin pill group,the ERK protein expression level,and the ERK mRNA amplification multiple were lower in the FOB group(P < 0. 01, P < 0. 05),and the MEK protein expression level had no significant difference(P > 0. 05). ConclusionThe FOB can promote the angiogenesis of myocardial infarction and increase the number of neovascularization at the edge of myocardial infarction. The mechanism may be related to the up-regulation of serum TGF- β level and the expression of the MEK/ERK signaling pathway.
