富勒烯 (C60) 对RNA逆转录的影响

摘要: 目的:研究富勒烯 (C60) 对RNA逆转录 (Reverse Transcription,RT) 反应的影响。方法:以斑马鱼鱼肝mRNA为模板,在M-MLV逆转录酶作用下合成cDNA,并在RT反应体系中添加不同浓度的C60,分别研究C60对M-MLV逆转录酶和RNA构象的影响。结果:100 μg/mL C60可显著抑制RT反应,抑制效应与C60浓度相关,但增加M-MLV逆转录酶量可以有效逆转C60对RT的抑制作用。C60与 RNA在37 ℃避光孵育12 h后,C60 可特异降解28S rRNA,同样具有浓度依赖性。结论:C60 不仅可显著抑制M-MLV逆转录酶活 性,而且对RNA也具有一定的损伤作用,C60具有潜在的基因毒性。

Abstract: Objective:To elucidate the effects of fullerene(C60)on reverse transcription(RT). Methods:Template mRNA was extracted from the liver of zebrafish,synthesised cDNA with M-MLV reverse transcriptase. Different concentrations of C60 were added into the RT reaction solution,to study the impact of C60 on M-MLV reverse transcriptase and RNA conformation respectively. Results:The results showed that 100 μg/mL of C60 could significantly inhibit the RT reaction,and this effect was related to the C60 concentration. The RT inhibition of C60 could be reversed by increasing the amount of M-MLV reverse transcriptase in the RT reaction solution. After its co-incubation with RNA in 37 ℃ for 12 h without light,C60 can specifically degraded 28S rRNA in the concentration-dependent manner. Conclusion:These results demonstrate that C60 could not only inhibit the activity of M-MLV reverse transcriptase,it also has damaging effect on the RNA. C60 may have potential genetics toxicity.
