摘要: 胰岛素样生长因子-1(IGF-1)是一种与生长激素产生生理作用过程息息相关的活性蛋白多肽物质,可以促进糖类、脂类、蛋白质和无机盐的代谢。然而,IGF-1长期使用给患者带来的不良反应大大限制了它的临床应用,为了扩大IGF-1在医学领域的应用,科研工作者正积极开发IGF-1相关的衍生物。综述了采用基因工程和分子生物学技术合成的截短型IGF-1、加长型IGF-1、IGF-1-Fc融合蛋白和聚乙二醇化的IGF-1衍生物,阐释了IGF-1衍生物优良的生物学功能,可为其他蛋白类药物的研发提供指导。

Abstract: Insulin-like growth factor-1(IGF-1)is an active protein polypeptide closely related to the physiological process of growth hormone,which can promote the metabolism of sugars,lipids,proteins and inorganic salts. In order to expand the application prospect of IGF-1 in the medical field,researchers are actively developing IGF-1 derivatives due to the adverse drug reaction brought by its long-term use,which greatly restrict its application. In this paper,the derivatives of truncated IGF-1,lengthened IGF-1,IGF-1-Fc fusion protein,pegylated IGF-1 synthesized by genetic engineering and molecular biology technology were reviewed,and the excellent biological functions of IGF-1 derivatives were explained,they provide guidance for the research and development of other protein drugs in the future.
