玉米杂交种瑞华玉288指纹图谱构建及其纯度鉴定

Construction of Maize Hybrid Ruihuayu 288 Fingerprint and Its Purity Identification

摘要: 为构建优良杂交玉米新品种瑞华玉288的指纹图谱,筛选适用于该品种纯度鉴定的InDel 引物,利用InDel分子标记技术构建指纹图谱,筛选引物对瑞华玉288种子进行纯度鉴定,并结合 田间小区种植鉴定法对鉴定结果进行验证。结果表明:在52对InDel引物中筛选出20对多态性 好、清晰稳定的引物构建瑞华玉288指纹图谱,多态性为38.46%;20对引物中有11对引物可以 用于玉米杂交种瑞华玉288纯度鉴定,利用其中的3对引物鉴定瑞华玉288纯度为96.26%,与田 间小区种植鉴定的结果98.13%高度一致,表明了分子鉴定法能够快速鉴定玉米杂交种的纯度, 有利于玉米种子质量的监控。

Abstract: To construct the fingerprint of the new hybrid corn variety Ruihuayu 288,the InDel primers suitable for its purity identification were screened. The fingerprint was constructed with InDel molecular marker technology,the purity of Ruihuayu 288 seeds was identified with the primers,and the identification results were verified by combining the field community planting identification method. The results showed that 20 pairs of primers with good polymorphism,clear,and stable primers were screened out of 52 pairs of InDel primers to construct the fingerprint of Ruihuayu 288,and the polymorphism was 38. 46%. Eleven pairs of primers in 20 pairs of primers could be used for the purity identification of maize hybrids Ruihuayu 288,and the purity of Ruihuayu 288 identified with 3 pairs of primers was 96. 26%,which was highly consistent with the results of 98. 13% of the field community planting identification. The results indicate that the molecular identification method can quickly identify the purity of maize hybrids, which is conducive to the monitoring of maize seed quality.
