藜蒿ISSR分子标记遗传多样性评价

Genetic Diversity Evaluation of ISSR Molecular Markers in Artemisia selengensis

摘要: 藜蒿是多年生草本植物,分布广泛,遗传背景复杂,系统分类模糊。为了解不同种群藜蒿 亲缘关系和遗传多样性,利用ISSR分子标记技术对我国9个省份不同地区的117份藜蒿种质资 源进行全面分析。通过9条ISSR引物检测到87个多态性位点数,多态性百分比为98.86%。不 同群体之间的Nei′s遗传多样性指数(H)平均值为0.099 6,Shannon 遗传多样性指数平均值为 0. 152 2。利用UPGMA 聚类分析117份藜蒿种质,遗传系数为0.35时分为3个主要类群。其中, 编号为41~ 44、110的5个样本归为第一类;其余的大多数样本归为第二类;编号47单独归为第 三类。表明不同藜蒿种质资源亲缘关系密切,遗传相似指数高。

Abstract: Artemisia selengensis is a perennial herb with a wide distribution,complex genetic background,and ambiguous systematic classification. The phylogenetic relationships of different populations of Artemisia selengensis should be understood to lay the foundation for genetic improvement. This study used ISSR molecular marker technology to comprehensively analyze 117 germplasm resources of Artemisia selengensis collected from different regions in 9 provinces of China. Using 9 ISSR primers,87 polymorphic loci were detected,with a polymorphism percentage of 98. 86%. The average Nei′s genetic diversity index(H)between different populations was 0. 099 6,and the average Shannon genetic diversity index was 0. 152 2. UPGMA cluster analysis grouped the 117 Artemisia selengensis samples into three main clusters at a genetic coefficient of 0. 35. Among them,most samples belonged to the second cluster,five samples numbered 41-44 and 110 were classified into the first cluster,and the sample numbered 47 formed the third cluster alone. The different germplasm resources of Artemisia selengensis have close phylogenetic relationships and high genetic similarity indices.
